The territory of Manzanares was originally populated by indigenous pantácoras and marquetones. During the Conquest, these Indians were subdued by the Spanish troops of Lieutenant Baltazar Maldonado, who was following orders of Gonzalo Jimenez de Quesada .

The district foundation is attributed to a group of settlers from Antioquia led by Alexander, Daniel and brothers Joaquín Marulanda. It was the seat of government of the state of Tolima for three months in the year 1867
